Personally, this was an incredibly complex paper to read though probably largely due to me being unfamiliar with many techniques and data analysis techniques (which were incredibly impressive) used in this paper. The introduction and theoretical background for this experiment were quite sound (though perhaps textually a bit too focused on neocortical layer 1 which does get top-down projections but Ndnf interneurons comprised only the third largest interneuron population of ZI-ACx projections and as far as I can tell they did not perturb this layer specifically) (this may be because the lab generally studies L1). I think this was a cool paper, it doesn't follow the "formula" of systems neuroscience research and uses methods and analysis techniques which are required by the questions they are asking (e.g., looking at synaptic boutons to study plasticity mechanisms). I need to reread this paper in detail to fully understand the technique in it. One star removed because the clarity of the text was sometimes lacking; complex ideas were presented but not well explained.
